Introduction

This meeting will address the spatiotemporal control of cellular events that proceeds through dynamically regulated multiprotein complexes. Information flow through these protein networks is finely tuned by the opposing activities of signal transduction and termination enzymes. The combination of new computational, proteomic, fluorescent imaging and structural biology techniques allows for analysis of how such dynamic protein networks propagate, process and disseminate second messenger signals at the molecular and cellular level. Moving from the concept of protein modules, the combinatorial assembly of multiprotein units is now explored. Cryoelectron microscopy and X-ray crystallography are used to define the structure of these higher order signalling complexes. Molecular imaging allows for deciphering the dynamic assembly of signalling complexes and second messenger signalling in situ. Molecular interaction techniques are exploited to make tools that can selectively perturb specific components of signal complexes. Single cell signal analyses combined with computational modeling can predict the ebb and flow of these signals within signal networks.
